263 SLFN11 is a possible biomarker of sensitivity to DNA-targeted regents in CD30-positive mycosis fungoides and Sezary syndrome

SLFN11 expressing tumor cells respond well to DNA-targeting anti-tumor reagents, as for its role in a dominant determination of sensitivity DNA damage responses. However, the roles patients with cutaneous T-cell lymphoma (CTCL) remain poorly understood. The aim this study is clarify expression profile major type CTCL, mycosis fungoides (MF) and Sezary syndrome (SS). Immunohistochemical showed that approximately half MF (13/25) 80% SS (4/5) were positive SLFN11. Interestingly, was significantly correlated both CD30 serum level soluble IL-2 receptor. In addition, co-expression detected some patients. Basically, T-cells lack steady state. we on normal induced upon CD3/CD28/IL-2 stimulation, resulting enhancement DNA-damaging agent, camptothecin. Among CTCL cell lines (Hut78, MJ, HH, MyLa, SeAx), Hut78 MJ expressed CD30, those suppressed by pan JAK inhibitor cerdulatinib. Furthermore, high-expressing sensitive etoposide, if compared other negative (HH, SeAx). Taken together, our findings suggested closely associated MF/SS, might be regulated via axis. We suppose predictive biomarker agents CD30-positive MF/SS.
